Christina Hawthorne (Jada Pinkett Smith) is het Hoofd medisch adviseur aan het Richmond Trinity Hospital. Dr. Tom Wakefield (Michael Vartan) werkt als hoofd chirurgie bij het zelfde ziekenhuis. Collega zusters zijn o.a. Bobbie Jackson (Suleka Mathew), Ray Stein (David Julian Hirsh) en Candy Sullivan (Christina Moore). Camille (Hannah Hodson) is Christina's dochter uit een multiculturele relatie en Amanda (Joanna Cassidy) is Christina’s schoonmoeder, welke toevallig ook een lid is van het bestuur van het ziekenhuis.
